Rat serum has a clearly defined role in research and diagnostics — not as a universal cell culture supplement, but as a species-specific control and blocking matrix.
In ELISA, Western blot, and immunofluorescence assays, rat serum is used as a blocking reagent when rat antibodies are used as primary or secondary antibodies. It prevents non-specific binding and reduces background without interfering with the target analyte. Typical concentration: 1–10% in PBS or TBS-Tween buffer.
When developing antibodies and diagnostic kits targeting rat proteins — or specifically designed to show no cross-reactivity with rats — rat serum is used as a negative control or competition reagent.
For the cultivation of rat cell lines (e.g. PC12, INS-1, NRK, H9c2, Rat-1) and rat primary cells (astrocytes, cardiomyocytes, hepatocytes), species-specific serum can improve growth conditions when a species-appropriate environment is required.
